| ARNT is a protein that plays a crucial role in the regulation of gene expression in response to environmental toxins and other stimuli. ARNT is a transcription factor that forms a heterodimer with other proteins, such as the aryl hydrocarbon receptor (AhR), to regulate the expression of target genes. ARNT is overexpressed in several types of cancer, including breast, lung, and colon cancer. In addition, ARNT has been shown to interact with other proteins that are involved in cancer development, such as the estrogen receptor and the androgen receptor. ARNT (Aryl Hydrocarbon Receptor Nuclear Translocator; HIF-1β) — Constitutively expressed basic helix-loop-helix PAS-family transcription factor that heterodimerizes with HIF-1α, HIF-2α, and related proteins to form functional hypoxia-inducible transcription complexes. ARNT is required for binding of HIF complexes to hypoxia-response elements (HREs) and subsequent regulation of genes involved in angiogenesis, glycolysis, oxygen adaptation, survival, and metabolism. ARNT also heterodimerizes with the aryl hydrocarbon receptor (AhR) and participates in xenobiotic-response signaling. Typical modulation in cancer: ↑ ARNT/HIF-1β activity can support tumor adaptation to hypoxia, angiogenesis, altered metabolism, survival, invasion, and treatment resistance when coupled to activated HIF-α signaling. However, ARNT is generally constitutively expressed and is less commonly the oxygen-regulated limiting component of the HIF pathway than HIF-1α. Cancer effects are therefore context-dependent. ARNT participates in HIF-mediated hypoxic responses and AhR signaling, both of which may influence oxidative stress, vascular function, inflammation, and neuronal adaptation, but there is no established universal AD-specific direction. |
