| WDR72 - WD Repeat Domain 72 Abbreviation: WDR72 Type: WD-repeat scaffold protein / protein-protein interaction regulator Function: WDR72 is a WD40 repeat-containing scaffold protein that provides a platform for assembly of protein complexes and is involved in regulation of cellular processes including proliferation, differentiation, adhesion, migration, vesicular trafficking, and tissue-specific functions such as enamel maturation. Cancer: ↑ WDR72 is overexpressed in several cancers and can promote tumor-cell proliferation, survival, migration, invasion, stemness, and metastasis. In esophageal squamous cell carcinoma, WDR72 activates PI3K/AKT/mTOR signaling and suppresses autophagy. In neuroblastoma, WDR72 promotes stemness in part by stabilizing CBX8 through interference with TRIM31-mediated degradation. Favorable Direction in Cancer: ↓ WDR72 expression or activity is generally favorable in tumors where WDR72 functions as an oncogenic scaffold and promotes PI3K/AKT/mTOR signaling, suppression of autophagy, proliferation, or stemness. Interpretation Note: WDR72 is not an enzyme, so changes should generally be recorded as expression or functional activity rather than catalytic inhibition. |
