Function: MMP16 is a membrane-associated matrix metalloproteinase that degrades extracellular-matrix components and can activate pro-MMP2. It regulates extracellular-matrix remodeling, cell migration, tissue invasion, and interactions between cells and the tumor microenvironment.

Cancer: ↑ MMP16 expression or activity is associated in several cancers with extracellular-matrix degradation, migration, invasion, metastasis, and tumor progression. MMP16 can promote invasive behavior directly through matrix proteolysis and indirectly through activation of other metalloproteinases such as MMP2.

Favorable Direction in Cancer: ↓ MMP16 expression or proteolytic activity is generally favorable where MMP16 promotes tumor invasion, extracellular-matrix remodeling, and metastasis.
