| CXCL16 - C-X-C Motif Chemokine Ligand 16 Abbreviation: CXCL16 Alternative Names: SR-PSOX, Scavenger Receptor for Phosphatidylserine and Oxidized LDL Type: Chemokine / membrane-associated cytokine / soluble chemotactic factor / scavenger receptor-like protein Function: CXCL16 exists as both a transmembrane protein and a soluble chemokine. It binds the receptor CXCR6 and regulates leukocyte recruitment, cell adhesion, migration, inflammatory signaling, and tissue immune responses. Membrane-associated CXCL16 can also function as a scavenger receptor for oxidized lipids. Cancer: ↑ CXCL16 expression or CXCL16-CXCR6 signaling is increased in several cancers and can promote tumor-cell proliferation, migration, invasion, angiogenesis, metastasis, and tumor-microenvironment signaling. However, CXCL16 can also recruit CXCR6-positive antitumor immune cells in some settings, making its net effect context-dependent. Favorable Direction in Cancer: ↓ CXCL16/CXCR6 signaling is generally favorable where the pathway directly promotes tumor growth, invasion, or metastasis. In immune-dominant contexts, increased CXCL16-mediated recruitment of cytotoxic lymphocytes may instead support antitumor immunity. Pathway Relationship: CXCL16 ↑ → CXCR6 activation ↑ → migration/invasion and downstream survival signaling ↑ in susceptible tumor cells. |
