| NR1D2 (Nuclear Receptor Subfamily 1 Group D Member 2; REV-ERBβ) — Nuclear receptor and transcriptional repressor that is a core component of the circadian clock. NR1D2/REV-ERBβ participates in transcriptional feedback with CLOCK/BMAL1 and regulates circadian rhythm, lipid and carbohydrate metabolism, inflammatory signaling, and other metabolic gene programs. It is closely related to NR1D1/REV-ERBα. Typical modulation in cancer: Context-dependent. REV-ERB receptors are frequently dysregulated in cancer and can influence proliferation, metabolism, survival, circadian control, and tumor-associated metabolic programs. Reduced REV-ERB expression has been reported in multiple cancers, although the biological effect varies by cancer type and experimental context. Typical modulation in Alzheimer’s disease: Potential relevance through circadian rhythm, metabolism, inflammation, and sleep regulation, but NR1D2 is not presently a core established Alzheimer’s disease target and no universal direction should be assigned. |
