| CS (Citrate Synthase) — Mitochondrial matrix enzyme that catalyzes the condensation of acetyl-CoA and oxaloacetate to form citrate, representing the first committed step of the tricarboxylic acid (TCA) cycle. Citrate synthase is widely used as a marker of mitochondrial oxidative capacity and mitochondrial content. Typical modulation in cancer: Context-dependent. Increased citrate synthase activity can support oxidative metabolism and TCA-cycle flux in tumors that rely on mitochondrial respiration, whereas reduced CS activity can indicate suppression of mitochondrial oxidative metabolism. Cancer cells differ substantially in their dependence on glycolysis versus oxidative phosphorylation. Typical modulation in Alzheimer’s disease: Reduced citrate synthase activity may reflect impaired mitochondrial function and reduced oxidative metabolism, both of which are relevant to neurodegenerative disease. |
