Type:

RRM2 (Ribonucleotide Reductase Regulatory Subunit M2) — Regulatory small subunit of ribonucleotide reductase, the enzyme responsible for conversion of ribonucleotides to deoxyribonucleotides (dNTPs). RRM2 is strongly cell-cycle regulated and is required for DNA synthesis, DNA replication, DNA repair, and maintenance of intracellular dNTP pools.

Typical modulation in cancer: ↑ RRM2 is frequently tumor-promoting and is associated with increased proliferation, DNA replication, invasion, treatment resistance, and poorer prognosis in many cancers. RRM2 overexpression can also contribute to resistance to nucleoside analog drugs such as gemcitabine by increasing competing intracellular dNTP pools. RRM2 inhibition or knockdown generally suppresses proliferation and can increase DNA damage and apoptosis.

Ferroptosis relevance: RRM2 can function as an endogenous ferroptosis suppressor in several cancer models. RRM2 supports glutathione-related antioxidant capacity and nucleotide metabolism; RRM2 inhibition or knockdown can increase lipid peroxidation and ferroptosis in susceptible tumor cells.
