| Alanine (Ala) — Nonessential amino acid produced primarily through transamination of pyruvate. Alanine participates in amino-acid metabolism, nitrogen transport, gluconeogenesis, and the glucose-alanine cycle and can serve as an alternative carbon source for cellular metabolism. Typical modulation in cancer: Context-dependent. Alanine can support tumor metabolism as a carbon and nitrogen source, particularly in metabolically stressed tumors. In pancreatic cancer, stromal cells can supply alanine to tumor cells, where it contributes carbon to the TCA cycle and supports biosynthesis and growth. Typical mechanistic relationships: Pyruvate ↔ alanine via ALT/GPT; alanine → pyruvate → TCA cycle / gluconeogenesis. |
