GNAI1 (G Protein Subunit Alpha I1) — Gene encoding the heterotrimeric G-protein alpha-i1 subunit (Gαi1), a member of the inhibitory Gi/o family of G proteins. GNAI1 transduces signals from multiple G-protein-coupled receptors and typically inhibits adenylyl cyclase, resulting in reduced intracellular cAMP and altered downstream PKA, MAPK, PI3K/AKT, ion-channel, and Gβγ-dependent signaling.

Typical modulation in cancer: Context-dependent. Altered GNAI1 expression or signaling can influence proliferation, migration, invasion, survival, and response to GPCR-mediated signals. Depending on tumor type and receptor context, GNAI1 may have either tumor-promoting or tumor-suppressive effects, so a universal ↑ or ↓ cancer direction should not be assigned.

Typical mechanistic relationship: Gi-coupled GPCR activation → GNAI1/Gαi1 activation → adenylyl cyclase inhibition → cAMP ↓ → PKA signaling ↓, with additional signaling through Gβγ, PI3K/AKT, MAPK, and ion-channel pathways.
