| PGF (Placental Growth Factor; PlGF) — Secreted member of the vascular endothelial growth factor (VEGF) family that promotes angiogenesis, endothelial-cell proliferation and migration, inflammatory-cell recruitment, and vascular remodeling. PGF signals primarily through FLT1/VEGFR1 and can also interact functionally with VEGF signaling pathways. Typical modulation in cancer: Frequently ↑ and generally unfavorable. Increased PGF/PlGF signaling can promote tumor angiogenesis, endothelial-cell migration, stromal and myeloid-cell recruitment, tumor growth, invasion, metastasis, and resistance to anti-angiogenic therapy in selected cancers. Typical mechanistic relationship: PGF/PlGF → FLT1/VEGFR1 activation → endothelial and myeloid-cell signaling → migration, angiogenesis, inflammation, and tumor vascularization ↑. Hypoxia relevance: PGF expression can increase in hypoxic and inflammatory tissues and contributes to pathological angiogenesis and vascular remodeling. |
