ABCC5 (ATP Binding Cassette Subfamily C Member 5; Multidrug Resistance-Associated Protein 5, MRP5) — ATP-dependent membrane efflux transporter of the ABC transporter family. ABCC5 transports endogenous organic anions, including cyclic nucleotides such as cGMP and cAMP, and can export selected nucleotide analogues and drug metabolites.

Typical modulation in cancer: Frequently ↑ in drug-resistant tumor cells and generally unfavorable when increased expression reduces intracellular exposure to anticancer agents. Elevated ABCC5/MRP5 can contribute to multidrug resistance, altered nucleotide signaling, and reduced therapeutic sensitivity.

Typical mechanistic relationship: ABCC5/MRP5 expression or activity ↑ → drug/metabolite efflux ↑ → intracellular drug concentration ↓ → treatment resistance ↑.

Drug-resistance relevance: ABCC5 has been associated with resistance to selected nucleoside and nucleotide analogues and other anticancer compounds, although substrate specificity differs from other MRP family members such as ABCC1/MRP1 and ABCC2/MRP2.

Physiologic relevance: ABCC5 also regulates extracellular and intracellular cyclic-nucleotide balance through transport of cGMP and cAMP and therefore has functions beyond drug resistance.
