| 8-oxo-2′-Deoxyguanosine Abbreviation: 8-oxo-dG, 8-oxodG, 8-OHdG Alternative Names: 8-Oxo-7,8-dihydro-2′-deoxyguanosine, 8-Hydroxy-2′-deoxyguanosine Type: Oxidative DNA damage biomarker / oxidized DNA nucleoside / DNA lesion Function: 8-oxo-dG is formed when reactive oxygen species oxidize guanine residues in DNA or the deoxyguanosine nucleotide pool. It is one of the most widely used biomarkers of oxidative DNA damage and can be measured in cellular DNA, tissue, blood, urine, and other biological samples. Cancer: ↑ 8-oxo-dG generally indicates increased oxidative DNA damage and is associated with mutagenesis, genomic instability, carcinogen exposure, and cancer development or progression. Favorable Direction in Cancer Prevention: ↓ 8-oxo-dG is generally favorable because it indicates reduced oxidative DNA damage. Anticancer Treatment Interpretation: Context-dependent. ↑ 8-oxo-dG within treated cancer cells or tumor tissue can indicate successful induction of oxidative DNA damage by an anticancer treatment, whereas chronic systemic or normal-tissue 8-oxo-dG ↑ is generally unfavorable. Pathway Relationship: ROS ↑ → guanine oxidation ↑ → 8-oxo-dG ↑ → oxidative DNA damage/mutagenic lesions ↑. Repair Relationship: OGG1 and associated base-excision-repair pathways remove oxidized guanine lesions from DNA. Increased urinary 8-oxo-dG can therefore reflect both oxidative damage and subsequent repair/excretion. Interpretation Note: Tissue DNA 8-oxo-dG and urinary 8-oxo-dG should not be interpreted identically. Tissue 8-oxo-dG reflects retained oxidative DNA lesions, whereas urinary 8-oxo-dG can reflect whole-body oxidative damage and repair/excretion. |
