| ERLIN1 (ER Lipid Raft Associated 1; Erlin-1) — Endoplasmic-reticulum membrane protein of the SPFH family that forms complexes with ERLIN2 and participates in ER-associated protein degradation (ERAD), including degradation of inositol 1,4,5-trisphosphate receptors (IP3R). ERLIN1 also regulates intracellular calcium signaling, cholesterol binding, SREBP signaling, and lipid homeostasis. Typical modulation in cancer: ↑ ERLIN1 may be tumor-promoting in some cancers. In colorectal cancer, ERLIN1 is upregulated in tumor tissue; ERLIN1 knockdown suppresses proliferation and migration and increases apoptosis, whereas ERLIN1 overexpression promotes tumor-cell growth and migration. Thus, ERLIN1 ↓ is generally interpreted as anticancer in this context. Typical modulation in Alzheimer’s disease: No well-established Alzheimer’s disease-associated direction. ERLIN1 may have indirect relevance through ER protein quality control, calcium homeostasis, lipid metabolism, and ER stress. |
