| IL1RN (Interleukin-1 Receptor Antagonist; IL-1Ra) — Endogenous anti-inflammatory cytokine that competitively binds the interleukin-1 receptor and inhibits signaling induced by IL-1α and IL-1β. IL1RN regulates acute and chronic inflammatory responses and limits excessive IL-1-mediated cytokine signaling. Typical modulation in cancer: Context-dependent but generally anti-inflammatory and potentially tumor-suppressive when IL-1 signaling is driving tumor progression. ↑ IL-1Ra can suppress IL-1-mediated angiogenesis, inflammatory signaling, invasion, and metastasis. Experimental IL-1Ra administration has reduced tumor growth, angiogenesis, and metastasis in some tumor models. However, effects depend on tumor type and immune context. Typical modulation in Alzheimer’s disease: ↑ IL-1Ra is generally considered anti-inflammatory and potentially protective by opposing excessive IL-1α/IL-1β signaling and neuroinflammation. However, circulating IL-1Ra can also rise as a compensatory response to systemic inflammation, so elevated measured levels do not necessarily indicate reduced disease activity. |
