| TMT1A (Thiol Methyltransferase 1A; METTL7A) — Methyltransferase-family enzyme with reported mRNA m6A methyltransferase and thiol S-methyltransferase activity. TMT1A/METTL7A is associated with the endoplasmic reticulum and lipid droplets and can influence RNA stability, lipid/cholesterol metabolism, drug metabolism, and cellular signaling.
Typical modulation in cancer: Context-dependent. METTL7A can act as a tumor suppressor in some cancers but can also promote treatment resistance or tumor-supportive signaling in others. In colorectal cancer, METTL7A is reduced and functions as a tumor suppressor by binding SREBP1/SCAP, limiting SREBP1 nuclear translocation, reducing cholesterol synthesis, and suppressing tumor progression. In contrast, increased METTL7A has been linked to resistance to thiol-containing HDAC inhibitors and to tumor-promoting DDR1/STAT3/CXCL5 signaling in some breast cancer models.
