CAPN9 (Calpain 9; Calpain-9) — Calcium-dependent cysteine protease of the calpain family, expressed predominantly in gastrointestinal tissues. CAPN9 participates in regulated proteolysis, epithelial differentiation, cytoskeletal and signaling-protein processing, and gastrointestinal tissue homeostasis.

Typical modulation in cancer: Context-dependent. CAPN9 has been implicated in gastric and other gastrointestinal cancers, where altered expression may affect proliferation, differentiation, invasion, and proteolytic signaling. Unlike CAPN1 and CAPN2, CAPN9 is relatively tissue-restricted and should not be assigned a universal oncogenic or tumor-suppressive direction.

Typical mechanistic relationship: Ca2+ signaling → CAPN9 activation → limited proteolysis of cellular substrates → altered signaling, cytoskeletal organization, differentiation, and survival.
