Function: PXR is a ligand-activated nuclear receptor expressed predominantly in the liver and intestine. It heterodimerizes with RXR and regulates genes involved in xenobiotic metabolism, drug transport, bile-acid homeostasis, lipid metabolism, glucose metabolism, inflammation, and detoxification. Major downstream targets include CYP3A4 and multiple drug-transport proteins.

Cancer: ↕ Context-dependent. PXR activation can promote cancer-cell survival, proliferation, invasion, metastasis, and chemotherapy resistance by inducing drug-metabolizing enzymes and efflux transporters and by modulating apoptosis, oxidative stress, and inflammatory signaling. However, PXR can also exert protective or tumor-suppressive effects in selected tissues and disease contexts, so its cancer direction should be interpreted by tumor type and ligand.

Favorable Direction in Cancer: Context-dependent. In tumors where PXR promotes drug resistance or tumor progression, ↓ PXR activity is generally favorable.
