| CD40 (CD40 Molecule; TNFRSF5) — Cell-surface member of the tumor necrosis factor receptor superfamily that functions as an immune costimulatory receptor. CD40 is expressed on B cells, dendritic cells, macrophages, endothelial cells, and various malignant cells and is activated primarily by CD40 ligand (CD40L/CD154). Typical modulation in cancer: Context-dependent. Activation of CD40 on antigen-presenting cells can enhance dendritic-cell maturation, antigen presentation, T-cell priming, macrophage activation, and antitumor immunity. Conversely, CD40 signaling in some tumor cells can promote survival, proliferation, inflammatory signaling, or resistance. Agonistic CD40 antibodies are under investigation as cancer immunotherapies. Typical modulation in Alzheimer’s disease / neuroinflammation: Often associated with inflammatory activation. CD40-CD40L signaling can promote microglial and immune-cell activation, cytokine production, and neuroinflammatory responses. Typical mechanistic relationship: CD40L/CD154 → CD40 → TRAF proteins → canonical/non-canonical NF-kB, MAPK and PI3K/AKT signaling → immune-cell activation, antigen presentation and inflammatory gene expression. |
