| CHRM1 - Cholinergic Receptor Muscarinic 1 Abbreviation: CHRM1, M1, M1 mAChR Alternative Names: Muscarinic Acetylcholine Receptor M1 Type: G protein-coupled receptor / muscarinic acetylcholine receptor / cholinergic signaling receptor Function: CHRM1 is a Gq/11-coupled muscarinic acetylcholine receptor expressed prominently in the central nervous system and in selected peripheral tissues. Activation stimulates phospholipase C, intracellular Ca2+ signaling, PKC, MAPK/ERK, and other downstream pathways involved in neuronal excitability, synaptic plasticity, cognition, secretion, proliferation, and cell survival. Alzheimer's Disease: ↓ / impaired M1 muscarinic signaling is associated with cholinergic dysfunction in Alzheimer's disease. CHRM1 activation can improve cholinergic neurotransmission and cognition and can promote non-amyloidogenic APP processing while reducing Aβ formation and tau phosphorylation in experimental models. Favorable Direction in Alzheimer's Disease: ↑ CHRM1 activation or restoration of normal M1 signaling is generally favorable. Cancer: ↕ Context-dependent. CHRM1 signaling can either suppress or promote tumor progression depending on tissue type. In pancreatic cancer models, CHRM1 activation suppresses tumor growth and cancer stemness, whereas in other cancers, including selected skin cancer models, CHRM1 signaling may support proliferation and survival. Favorable Direction in Cancer: Context-dependent and should be assigned according to tumor type and experimentally observed signaling response. |
