| PROK2 (Prokineticin 2; PK2/Bv8) — Secreted prokineticin-family signaling protein and ligand for the G-protein-coupled receptors PROKR1 and PROKR2. PROK2 regulates angiogenesis, endothelial-cell proliferation, chemotaxis, circadian signaling, neurogenesis, inflammation, and smooth-muscle activity. Typical modulation in cancer: ↑ PROK2 is generally tumor-promoting in cancers where the PROK2 pathway is active, particularly through increased angiogenesis, recruitment of pro-angiogenic myeloid cells, migration, and tumor-supporting inflammatory signaling. PROK2 inhibition is therefore generally considered anticancer in these contexts. Typical modulation in Alzheimer’s disease: Context-dependent. PROK2 participates in circadian and neuronal signaling and can be induced by cellular stress, but there is not a sufficiently established universal AD direction. |
