| THBD (Thrombomodulin; CD141) — Endothelial cell-surface glycoprotein receptor that binds thrombin and alters its substrate specificity, promoting activation of protein C. Activated protein C subsequently inactivates coagulation factors Va and VIIIa, reducing further thrombin generation. THBD also contributes to endothelial protection and regulation of inflammatory and coagulation pathways. Typical modulation in cancer: Context-dependent. Loss or reduction of endothelial or tumor-cell THBD can be associated with increased coagulation, inflammation, invasion, and poorer prognosis in some cancers, whereas preserved or increased THBD may reflect a more differentiated or less aggressive phenotype. Effects vary substantially by tumor type. Typical modulation in Alzheimer’s disease: No established core Alzheimer’s disease-specific direction. THBD may have indirect relevance through endothelial dysfunction, vascular inflammation, coagulation, and blood-brain barrier-related mechanisms. |
