| NR1H4 (Nuclear Receptor Subfamily 1 Group H Member 4; Farnesoid X Receptor, FXR) — Ligand-activated nuclear receptor and transcription factor that functions primarily as a bile-acid sensor. FXR regulates bile-acid synthesis and transport, cholesterol homeostasis, lipid and glucose metabolism, enterohepatic signaling, inflammation, and hepatic metabolic gene expression. Typical modulation in cancer: Context-dependent. FXR can function as a tumor suppressor in some gastrointestinal and hepatic cancers by limiting inflammation, bile-acid toxicity, and proliferative signaling, whereas FXR activation may support survival or metabolic adaptation in selected tumor contexts. A universal ↑ or ↓ cancer direction should therefore not be assigned. Typical mechanistic relationships: Bile acids → FXR activation → transcriptional regulation of genes involved in bile-acid synthesis/transport, cholesterol metabolism, lipid metabolism, and inflammatory signaling. abbreviations: NR1H4; common name: FXR; aliases include BAR, RIP14, HRR1. |
