miR-375 is a regulatory microRNA that generally functions as a tumour suppressor in head-and-neck and HPV-associated squamous cell carcinomas. It can suppress oncogenic transcripts involved in proliferation, survival, invasion, and extracellular-matrix degradation, including AEG-1/MTDH, IGF1R, JAK2, and YAP1. miR-375 may also directly or indirectly reduce HPV16/18 E6 and E7 expression, thereby supporting p53 and RB tumour- suppressor activity. miR-375 is frequently downregulated in these cancers; therefore, restoration or increased expression is generally considered the favourable direction in this context. Effects may differ by cancer type.
