FEN1 Flap endonuclease 1 DNA replication and repair Up Structure-specific nuclease involved in Okazaki-fragment maturation, long-patch base-excision repair and replication-fork processing. FEN1 is frequently overexpressed in cancer and can promote tumour-cell proliferation, DNA-damage repair and treatment resistance. Therapeutic inhibition or downregulation increases replication stress and DNA damage, suppresses proliferation and may produce synthetic lethality in homologous-recombination-deficient cancers.
