| FcεRI - High-Affinity Immunoglobulin E Receptor Abbreviation: FcεRI, Fc epsilon RI Canonical IgE-Binding Subunit: FCER1A / FcεRIα Type: High-affinity IgE receptor / immune-cell activation receptor Function: FcεRI is the high-affinity receptor for immunoglobulin E (IgE) and is expressed prominently on mast cells and basophils, with expression also occurring on selected antigen-presenting cells. IgE binds the FcεRIα subunit, sensitizing the cell to specific allergens. Allergen-mediated cross-linking of FcεRI-bound IgE activates intracellular signaling involving Lyn, Fyn, Syk, PLCγ, PI3K, calcium signaling, and NF-κB, leading to mast-cell or basophil degranulation and production of lipid mediators and cytokines. Asthma: ↑ Increased FcεRI expression and/or activation contributes strongly to allergic asthma. Elevated IgE stabilizes FcεRI on the surface of mast cells and basophils, increasing sensitivity to allergens. Allergen-induced FcεRI activation causes release of histamine, tryptase, leukotrienes including LTC4, prostaglandin D2, cytokines, and chemokines, promoting bronchoconstriction, mucus secretion, airway inflammation, airway hyperresponsiveness, and remodeling. Reduced FcεRI signaling is generally favorable in allergic asthma. Favorable Direction in Asthma: ↓ FcεRI expression or activation reduces IgE-mediated mast-cell and basophil activation and allergic airway inflammation. |
