HTRA1 is a secreted/extracellular serine protease involved in protein quality control, extracellular matrix remodeling, TGF-β-family signaling, and disease-related proteolysis. The human HtrA family has four members, HtrA1–4, and reviews link HtrA dysregulation to cancer, arthritis, neurodegenerative disease, and reproductive disorders.

-most literature supports HTRA1 and HTRA3 as tumor-suppressive, while HTRA2 is more often associated with tumor growth and metastasis.
