AFAP1 antisense RNA 1 (AFAP1-AS1; also called AFAP1AS or AFAP1-AS) is a long non-coding RNA transcribed antisense to the protein-coding AFAP1 locus. AFAP1-AS1 regulates gene expression through mechanisms that can include microRNA sequestration, interaction with regulatory proteins, modulation of mRNA stability, and epigenetic control of transcription. In many solid cancers, AFAP1-AS1 is the predominant pattern and is associated with increased proliferation, survival, migration, invasion, epithelial-mesenchymal transition, metastasis, treatment resistance, and poorer prognosis. Conversely, AFAP1-AS1 knockdown or expression ↓ commonly reduces tumour-cell growth and motility and can increase apoptosis or treatment sensitivity in experimental models. Reported downstream pathways and targets include c-MYC, WNT/β-catenin, PI3K/AKT, RAS/MEK, EMT-related transcription factors, EZH2-mediated gene repression, and multiple tumour-specific microRNA axes.
