SRC-3 (steroid receptor coactivator-3; NCOA3; AIB1) is a transcriptional coactivator encoded by the NCOA3 gene. It does not function as a steroid receptor itself; instead, it binds activated nuclear receptors and other transcription factors and enhances expression of genes involved in proliferation, metabolism, survival, migration and invasion. SRC-3 is frequently amplified, overexpressed or excessively activated in cancer, particularly breast cancer, and can promote estrogen-receptor signaling, PI3K/AKT signaling, growth-factor responses, epithelial–mesenchymal transition, metastasis and resistance to endocrine or anticancer therapy. The typical cancer-associated direction is up, while the desired anticancer modulation is generally down, functional inhibition or protein degradation.
