| XPF - ERCC Excision Repair 4 Endonuclease Type: Structure-specific DNA repair endonuclease / nucleotide excision repair protein Function: XPF forms a heterodimer with ERCC1 to generate the ERCC1-XPF structure-specific endonuclease. The complex performs the 5' incision during nucleotide excision repair and also participates in repair of DNA interstrand crosslinks, double-strand breaks, and other structurally complex DNA lesions. Cancer: ↑ Increased ERCC1-XPF DNA-repair activity can promote resistance to platinum-based and other DNA-damaging anticancer therapies by repairing treatment-induced DNA lesions. Pharmacologic or genetic inhibition of XPF/ERCC1 can increase tumor-cell sensitivity to cisplatin. However, loss of XPF-mediated DNA repair can also increase genomic instability and cancer susceptibility, so its role differs between cancer initiation and treatment of established tumors. |
