ABCG1 - ATP-Binding Cassette Subfamily G Member 1

Type: ATP-binding cassette lipid transporter / cholesterol efflux transporter

Function: ABCG1 is an ATP-dependent membrane transporter that regulates intracellular cholesterol and phospholipid homeostasis, particularly by promoting cholesterol efflux to mature HDL particles. ABCG1 influences membrane lipid composition, lipid-raft signaling, macrophage function, cellular metabolism, and inflammatory responses.

Cancer: ↕ Context-dependent, with a predominantly tumor-promoting pattern in many cancers. Elevated ABCG1 can support cancer-cell survival, stem-like properties, proliferation, chemoresistance, Akt-associated signaling, and tumor-promoting macrophage polarization. However, in some cancer models increased ABCG1-mediated cholesterol efflux depletes cellular cholesterol and suppresses proliferation or promotes apoptosis.

Alzheimer's Disease: ↕ Context-dependent. ABCG1 regulates cholesterol efflux and membrane cholesterol distribution and can influence α-, β-, and γ-secretase activity and Aβ production. Experimental studies report both increased and decreased Aβ following ABCG1 modulation, so a consistent disease-associated direction has not been established.
