| CYP4A - Cytochrome P450 Family 4 Subfamily A Human Members: CYP4A11, CYP4A22 Type: Cytochrome P450 monooxygenase subfamily / fatty-acid ω-hydroxylases Function: CYP4A enzymes catalyze ω-hydroxylation of fatty acids and participate in the metabolism of arachidonic acid and other lipids. CYP4A-dependent arachidonic-acid metabolism contributes to production of 20-hydroxyeicosatetraenoic acid (20-HETE), a bioactive lipid mediator involved in vascular regulation, inflammation, cellular proliferation, and angiogenic signaling. In humans, CYP4A11 is an important CYP4A enzyme involved in 20-HETE synthesis, while CYP4A22 is a closely related family member. Cancer: ↑ Increased CYP4A/20-HETE pathway activity is generally associated with tumor-promoting effects. CYP4A-derived 20-HETE can stimulate cancer-cell proliferation, survival, angiogenesis, migration, and tumor growth. CYP4A/4F expression is elevated in several tumor types, and pharmacological inhibition of 20-HETE-producing CYP enzymes suppresses growth in multiple experimental cancer models. |
