CTSK - Cathepsin K

Type: Lysosomal cysteine protease / collagenase

Function: Cathepsin K is a papain-family lysosomal cysteine protease with potent collagenolytic and elastolytic activity. It is highly expressed in osteoclasts and plays a major role in degradation of type I collagen and other extracellular-matrix proteins during bone resorption. CTSK also participates in extracellular-matrix remodeling, inflammatory signaling, epithelial-mesenchymal transition, and interactions between tumor cells and the bone microenvironment.

Cancer: ↑ Frequently increased or activated in cancer cells, osteoclasts, and cells of the tumor microenvironment. Elevated CTSK promotes extracellular-matrix degradation, tumor-cell migration and invasion, epithelial-mesenchymal transition, angiogenesis, osteolytic bone destruction, and establishment of bone metastases. CTSK inhibition can suppress invasion, bone resorption, and tumor progression in preclinical models.
