Function: 3-Nitrotyrosine is formed by nitration of tyrosine residues in proteins, primarily through reactive nitrogen species such as peroxynitrite. It is widely used as a biomarker of nitrosative stress, oxidative protein damage, and excessive nitric oxide/reactive oxygen species interactions. Protein tyrosine nitration can alter enzyme activity, protein structure, signaling, and cellular function.

Cancer: ↑ Frequently increased in tumors and tumor-associated inflammatory environments. Elevated nitrotyrosine reflects increased reactive oxygen and nitrogen species, peroxynitrite formation, inflammation, and oxidative protein damage and can accompany tumor progression, angiogenesis, immune dysfunction, and treatment-related oxidative stress.

Alzheimer's Disease: ↑ Increased 3-nitrotyrosine and protein tyrosine nitration are well-established indicators of oxidative and nitrosative stress in Alzheimer's disease. Elevated nitration affects proteins involved in energy metabolism, mitochondrial function, cytoskeletal regulation, and synaptic function and is associated with neuronal dysfunction and neurodegeneration.
