| CYP2C8 - Cytochrome P450 Family 2 Subfamily C Member 8 Abbreviation: CYP2C8 Type: Cytochrome P450 monooxygenase / drug-metabolizing enzyme / xenobiotic metabolism enzyme Function: CYP2C8 is an endoplasmic-reticulum cytochrome P450 enzyme expressed predominantly in the liver. It catalyzes oxidative metabolism of numerous drugs and xenobiotics and also metabolizes endogenous compounds including arachidonic acid and retinoids. Drug and Compound Metabolism: ↕ CYP2C8 activity can increase or decrease the effective exposure to a compound depending on whether metabolism inactivates the parent compound, generates an active metabolite, or contributes to metabolic clearance. Cancer: ↕ Context-dependent. CYP2C8 metabolizes several anticancer and cancer-relevant compounds, including paclitaxel. Increased CYP2C8 activity can increase metabolic clearance of certain drugs, whereas CYP2C8 inhibition can increase their systemic exposure. The biological consequence therefore depends on the substrate and treatment context. Pharmacogenetics: CYP2C8 has functional genetic variants that can alter enzyme activity and drug clearance, contributing to interindividual differences in pharmacokinetics and treatment response. Interpretation Note: When a natural compound inhibits CYP2C8, record CYP2C8 activity ↓. Do not automatically interpret this as favorable or unfavorable, because inhibition may increase exposure to CYP2C8 substrates and therefore increase either efficacy or toxicity. |
