| PRMT6 - Protein Arginine Methyltransferase 6 Abbreviation: PRMT6 Type: Type I protein arginine methyltransferase / epigenetic regulator / histone methyltransferase Function: PRMT6 catalyzes asymmetric dimethylation of arginine residues on histone and non-histone proteins. A major chromatin mark produced by PRMT6 is H3R2me2a, which can alter transcription through crosstalk with other histone modifications such as H3K4me3. PRMT6 also regulates cell-cycle proteins, signaling proteins, RNA-processing factors, and metabolic enzymes. Cancer: ↕ Context-dependent. PRMT6 is oncogenic in many cancers, where increased expression or activity can promote proliferation, migration, invasion, epithelial-to-mesenchymal transition, stemness, metabolic reprogramming, and treatment resistance. However, PRMT6 can also act as a tumor suppressor in selected contexts, including hepatocellular carcinoma and melanoma. Favorable Direction in Cancer: Context-dependent. ↓ PRMT6 may be favorable in tumors where PRMT6 drives oncogenic transcription or signaling, whereas ↑ PRMT6 may be favorable where it suppresses pathways such as RAS-MEK/ERK or represses tumor-promoting genes. Interpretation Note: PRMT6 direction should be assigned according to tumor type and substrate. H3R2me2a ↑ alone should not automatically be interpreted as favorable or unfavorable because PRMT6 can repress either tumor-suppressor or oncogenic genes depending on chromatin context. |
