GSTP1/GSTπ Cancer Research Results

GSTP1/GSTπ, Glutathione S-transferase pi 1: Click to Expand ⟱
Source:
Type:
An enzyme that plays a crucial role in the detoxification of various substances, including carcinogens. It is also involved in the regulation of cellular processes, such as cell growth, differentiation, and apoptosis.
GSTP1 is overexpressed in various types of cancer, including prostate, breast, lung, and colon cancer. The overexpression of GSTP1 has been associated with cancer progression, metastasis, and poor prognosis.

265- ALA,    Alpha-Lipoic Acid Reduces Cell Growth, Inhibits Autophagy, and Counteracts Prostate Cancer Cell Migration and Invasion: Evidence from In Vitro Studies
- in-vitro, Pca, LNCaP - in-vitro, Pca, DU145
ROS↓, ALA decreased ROS production, SOD1 and GSTP1 protein expression
SOD↓, SOD1, DU145
GSTP1/GSTπ↓,
NRF2↓, significantly reduced the cytosolic and nuclear content of the transcription factor Nrf2
p62↓, du145
p62↑, LNCaP
SOD↑, LNCaP
p‑mTOR↑, revealed that in both cancer cells, ALA, by upregulating pmTOR expression, reduced the protein content of two autophagy initiation markers, Beclin-1 and MAPLC3.
Beclin-1↓,
ROS↑, Interestingly, in LNCaP cells, we observed an almost significant increase in ROS content (p = 0.06) after ALA compared to the control, concomitantly with a significant upregulation of the antioxidant enzyme SOD1 after 48 h.
SOD1↑,

576- ART/DHA,    Profiling of Multiple Targets of Artemisinin Activated by Hemin in Cancer Cell Proteome
- Analysis, NA, NA
GSTP1/GSTπ↓, IC50 value of 2 μM
TfR1/CD71↓,

6075- CHL,  docx,    The effect of the combination therapy with chlorophyllin, a glutathione transferase P1-1 inhibitor, and docetaxel on triple-negative breast cancer invasion and metastasis in vivo/in vitro
- vitro+vivo, BC, 4T1
TumCMig↓, the coadministration of chlorophyllin and docetaxel significantly inhibited cell migration in vitro.
eff↑, coadministration of chlorophyllin and docetaxel may have a potential role in controlling metastatic processes by suppressing cell migration, gelatinase activity, and micrometastasis formation in triple-negative breast cancers.
TumMeta↓,
TumCCA↑, We found that while the docetaxel decreased the G0/G1 phase, both chlorophyllin and its coadministration with docetaxel led to a slight increase in the G0/G1 phase
Trx↓, In a study, it is shown that chlorophyllin inhibits the cellular thioredoxin reductase activity in a time-dependent manner in several cancer cell lines, induces ROS accumulation, and leads to cell death
ROS↓,
TumCD↑,
GSTP1/GSTπ↓, Chlorophyllin was shown to be a specific inhibitor of GSTP1.

441- CUR,    Curcumin Regulates ERCC1 Expression and Enhances Oxaliplatin Sensitivity in Resistant Colorectal Cancer Cells through Its Effects on miR-409-3p
- in-vitro, CRC, HCT116
ERCC1↓,
Bcl-2↓,
GSTP1/GSTπ↓,
MRP↓,
P-gp↓,
miR-409-3p↑,
survivin↓,

1941- PL,    Piperlongumine selectively kills cancer cells and increases cisplatin antitumor activity in head and neck cancer
- in-vitro, HNSCC, NA
selectivity↑, Piperlongumine killed HNC cells regardless of p53 mutational status but spared normal cells.
eff↑, Piperlongumine increased cisplatin-induced cytotoxicity in HNC cells in a synergistic manner in vitro and in vivo.
ROS↑, Piperlongumine selectively increases ROS accumulation in HNC cells
toxicity↑, PL markedly induced death in cancer cells, while the viability of normal cells was affected only minimally at the highest concentration (15 μM) tested
GSH↓, PL decreased GSH levels and increased GSSG levels in HNC cells (Figure 2 and Supplementary Figure S1); however, PL did not increase GSSG levels in normal HOK-1 cells
GSSG↑,
*GSSG∅, however, PL did not increase GSSG levels in normal HOK-1 cells
cl‑PARP↑, PL increased the levels of PARP and PUMA proteins regardless of p53 status
PUMA↑,
GSTP1/GSTπ↓, PL regulates ROS by targeting GSTP1, a direct negative regulator of JNK [22, 23], and thereby increases JNK phosphorylation
ChemoSen↑, Piperlongumine increases the cytotoxicity of cisplatin in HNC cells in vitro and in vivo

2951- PL,  AF,    Synergistic Dual Targeting of Thioredoxin and Glutathione Systems Irrespective of p53 in Glioblastoma Stem Cells
- in-vitro, GBM, U87MG
GSH↓, natural product piperlongumine (PPL) inhibit the GSH system.
eff↑, subsequent efficacy of PLL in combination with Au within a nanomolar range in GSCs. Auranofin (Au), a known TrxR1 inhibitor
GSTP1/GSTπ↓, PPL is a natural alkaloid found in the plant Piper longum Linn that directly inhibits GSTP-1 and exhibits anti-cancer properties

4730- Se,    Association between plasma selenium level and NRF2 target genes expression in humans
- Human, Nor, NA
*NRF2↑, NRF2 mRNA level was positively correlated with expression of investigated NRF2-target genes.
*GSTP1/GSTπ↓, plasma Se level was significantly inversely associated only with expression of GSTP1 (β-coef. = −0.270, p = 0.009), PRDXR1 (β-coef. = −0.245, p = 0.017) and SOD2 with an inverse trend toward significance
*SOD2↓,
