Vacuole membrane protein 1 (VMP1) is a
transmembrane protein that plays an important role in autophagosome formation,
autophagic flux, membrane organization, and endoplasmic-reticulum contact
sites. In cancer, VMP1 expression and activity are
context-dependent. In many established tumours,
VMP1 ↑ can increase protective autophagy, allowing
cancer cells to survive nutrient deprivation, hypoxia, oxidative stress, and
anticancer treatment. VMP1-mediated autophagy has been associated with tumour
progression and chemotherapy resistance, particularly in pancreatic cancer,
where KRAS signalling and gemcitabine treatment can induce VMP1. Conversely,
deficient VMP1 or impaired autophagic flux may contribute to cellular damage
and tumour initiation in selected settings.
