PGK2 (phosphoglycerate kinase 2) is a glycolytic enzyme that catalyzes the reversible conversion of 1,3-bisphosphoglycerate to 3-phosphoglycerate while generating ATP. PGK2 is normally expressed primarily in male germ cells and functions as a testis-specific counterpart of the ubiquitously expressed PGK1. Aberrant PGK2 expression has been reported in some cancers and may reflect activation of germ-cell or cancer-testis transcriptional programs, but its direct contribution to tumour metabolism and progression is substantially less established than that of PGK1. Where tumour-cell PGK2 overexpression is demonstrated, it could increase glycolytic ATP production and support proliferation; however, current evidence does not justify assigning PGK2 a universal cancer-associated direction. Therefore, its cancer modulation should be considered context-dependent and recorded from the individual study.
