Type:

LPAR1 - Lysophosphatidic Acid Receptor 1

Abbreviation: LPAR1, LPA1, EDG2

Type: G protein-coupled receptor (GPCR) / lysophosphatidic acid receptor

Function: Cell-surface receptor for lysophosphatidic acid (LPA) that regulates cell survival, proliferation, migration, cytoskeletal remodeling, adhesion, and invasion through signaling pathways including RhoA/ROCK, PI3K/AKT, MAPK, and related downstream pathways.

Cancer: ↑ Frequently increased or overactivated in tumor progression. LPA-LPAR1 signaling can promote proliferation, migration, invasion, epithelial-mesenchymal transition, angiogenic signaling, and metastasis. Increased LPAR1 expression has been associated with more invasive or metastatic phenotypes in several cancers, while LPAR1 inhibition or knockdown can suppress LPA-induced invasion and metastatic behavior.

- vitro+vivo, AD, SH-SY5Y
*MAPK↓, down-regulated the p38 MAPK, NF-κB p65, and STAT3 signaling pathways.
*NF-kB↓,
*STAT3↓,
*ROS↓, KRGM-G also prevented the formation of reactive oxygen species and stimulated the Nrf2-HO-1/4-HNE signaling pathway in the brains of [Formula: see text] mice and SH-SY5Y cells.
*NRF2↑,
*HO-1↑,
*HNE↑,
*LPAR1/EDG2↑, associated with up-regulation of LPAR1 in the brains of [Formula: see text] mice.
*cognitive↑, results suggest that KRGM-G might improve AD-related cognitive dysfunction by stimulating the anti-oxidant pathway (Nrf2) and inhibiting inflammatory pathways (p38/NF-κB/STAT3) through LPAR1.
