| IGF-2R - Insulin-Like Growth Factor 2 Receptor Abbreviation: IGF-2R, IGF2R, CI-M6PR, M6P/IGF2R Type: Growth-factor scavenger receptor / cation-independent mannose-6-phosphate receptor Function: Cell-surface and intracellular trafficking receptor that binds insulin-like growth factor 2 (IGF-2) and mannose-6-phosphate-containing proteins. IGF2R internalizes and directs IGF-2 toward lysosomal degradation, thereby limiting IGF-2-mediated mitogenic signaling. It also regulates lysosomal enzyme trafficking and contributes to activation of TGF-β. Cancer: ↓ Frequently reduced, mutated, or lost in cancer and generally functions as a tumor suppressor. Loss of IGF2R can increase extracellular IGF-2 availability and enhance proliferative IGF signaling. Loss of heterozygosity and inactivating alterations of IGF2R have been reported in multiple malignancies and can be associated with tumor progression and poorer outcome. |
