TRADD - TNF Receptor Type 1-Associated Death Domain Protein

Type: Death-domain adaptor protein / TNF receptor signaling protein

Function: TRADD is a signaling adaptor recruited primarily to TNFR1 and other death-domain-containing receptors. It provides a platform for recruitment of proteins including TRAF2, RIPK1, and FADD and can regulate NF-κB and MAPK activation, inflammation, apoptosis, necroptosis, cell survival, and proliferation.

Cancer: ↕ Context-dependent. TRADD can promote tumor-suppressive death-receptor signaling through recruitment of FADD and activation of caspase-mediated apoptosis. However, TRADD can also activate NF-κB, MAPK, and inflammatory survival pathways that support proliferation and tumor-cell survival. Its net cancer effect therefore depends on receptor, cellular context, and downstream signaling-complex formation.
