TRAF2 - TNF Receptor Associated Factor 2

Type: Signaling adaptor / E3 ubiquitin ligase-associated protein

Function: TRAF2 is a signaling adaptor recruited by multiple TNF receptor superfamily members and forms complexes with proteins including cIAP1/BIRC2, cIAP2/BIRC3, RIPK1, and other signaling regulators. It controls NF-κB, JNK/MAPK, inflammatory, survival, apoptotic, and immune-signaling pathways through ubiquitination-dependent signaling complexes.

Cancer: ↑ Frequently increased or functionally overactive in cancer. TRAF2 can promote NF-κB and MAPK signaling, tumor-cell survival, proliferation, inflammation, resistance to apoptosis, and treatment resistance. Amplification or increased TRAF2 signaling has been reported in several malignancies, although its effects depend on receptor and downstream signaling context.
