Type: Tight-junction transmembrane protein / epithelial adhesion and barrier protein

Function: Claudin-7 is a four-pass transmembrane protein of epithelial tight junctions that helps regulate paracellular permeability, epithelial polarity, cell-cell adhesion, and tissue barrier integrity. CLDN7 also influences epithelial-mesenchymal transition, migration, invasion, and signaling pathways including Wnt/β-catenin and adhesion-related signaling.

Cancer: ↕ Context-dependent. Loss or reduced CLDN7 expression occurs in several cancers and can disrupt epithelial adhesion, reduce E-cadherin, promote EMT, and increase invasion and metastasis. Conversely, CLDN7 is overexpressed in other tumors, including subsets of ovarian, gastric, pancreatic, prostate, and breast cancers, where increased or abnormally localized CLDN7 can promote migration, invasion, metastasis, and treatment resistance. Interpretation should therefore be cancer-type and localization specific.
