Function: ACADS encodes short-chain acyl-CoA dehydrogenase, a mitochondrial flavoprotein that catalyzes the first dehydrogenation step in β-oxidation of short-chain fatty acyl-CoAs, particularly butyryl-CoA. ACADS contributes to fatty-acid energy metabolism, mitochondrial function, and metabolic homeostasis.

Cancer: ↓ Reduced ACADS expression has been reported in cancers including colorectal cancer and hepatocellular carcinoma and is associated with tumor progression and poorer prognosis. Experimental restoration of ACADS can suppress proliferation, migration, and invasion. ACADS may also promote antitumor immunity by enhancing mitochondrial DNA leakage and cGAS-STING signaling, while ACADS deficiency can contribute to an immunosuppressive tumor microenvironment.
