EDEM Family - ER Degradation-Enhancing α-Mannosidase-Like Proteins

Members: EDEM1, EDEM2, EDEM3

Type: ER-associated degradation regulators / glycoprotein quality-control proteins / GH47 family

Function: EDEM proteins participate in endoplasmic-reticulum-associated degradation (ERAD) by recognizing and processing terminally misfolded glycoproteins and promoting their removal from the ER for proteasomal degradation. EDEM proteins are induced during ER stress and form part of the unfolded protein response-associated protein quality-control system.

Cancer: ↕ Context-dependent. Increased EDEM/ERAD activity can help tumor cells survive chronic proteotoxic, metabolic, and hypoxic stress by clearing misfolded proteins, while excessive or disrupted ERAD can contribute to ER stress and cell death. Cancer effects therefore depend on tumor type and ER-stress context.
