ULK1 - Unc-51 Like Autophagy Activating Kinase 1

Type: Serine/threonine kinase / autophagy initiation regulator

Function: ULK1 is a core kinase that initiates autophagy as part of the ULK1-ATG13-RB1CC1/FIP200-ATG101 complex. Under nutrient or energy stress, ULK1 activates downstream autophagy machinery and promotes phagophore nucleation and autophagosome formation. ULK1 is inhibited by mTORC1 and activated through nutrient- and energy-sensing pathways including AMPK.

Cancer: ↕ Context-dependent. ULK1-mediated autophagy can suppress early tumor development by maintaining cellular quality control and removing damaged organelles, but established tumors can exploit ULK1-dependent autophagy to survive hypoxia, nutrient deprivation, metabolic stress, detachment, and anticancer treatment.

Favorable Direction in Cancer: Context-dependent. ↓ ULK1 activity may be favorable in autophagy-dependent established tumors, whereas loss of normal ULK1-mediated quality control can potentially facilitate tumor initiation.

Alzheimer's Disease: ↓ ULK1 expression/activity has been associated with impaired autophagy and mitophagy in Alzheimer's disease. Reduced ULK1 can contribute to defective clearance of damaged mitochondria and pathological proteins such as Aβ and tau.

Favorable Direction in Alzheimer's Disease: ↑ Restoration or activation of normal ULK1-dependent autophagy and mitophagy is generally favorable where ULK1 activity is impaired.

Interpretation Note: ULK1 is the main mammalian target corresponding to the older ATG1 designation. ULK2 is a related paralog and should be recorded separately when specifically measured.
