Cellular senescence is a state in which cells cease to divide and grow, often in response to DNA damage or other forms of cellular stress.
On one hand, senescence can prevent cancer by:
-Halting the proliferation of damaged cells, thereby preventing the accumulation of mutations that can lead to cancer
-Activating immune responses that can eliminate senescent cells.
-Producing anti-tumor factors that can inhibit the growth of nearby cancer cells.

On the other hand, senescence can also promote cancer by:
-Creating a pro-tumorigenic microenvironment that can support the growth of nearby cancer cells
-Producing pro-inflammatory factors that can promote cancer progression
-Contributing to the development of a senescence-associated secretory phenotype (SASP), which can promote cancer cell growth and metastasis.

7411- CS,    Long Term Exposure to Polyphenols of Artichoke (Cynara scolymus L.) Exerts Induction of Senescence Driven Growth Arrest in the MDA-MB231 Human Breast Cancer Cell Line
- in-vitro, BC, MDA-MB-231 - in-vitro, CRC, HCT116
AntiCan↑, Polyphenolic extracts from the edible part of artichoke (Cynara scolymus L.) have been shown to be potential chemopreventive and anticancer dietary compounds.
chemoPv↑,
Apoptosis↑, High doses of polyphenolic extracts (AEs) induce apoptosis and decrease the invasive potential of the human breast cancer cell line, MDA-MB231.
TumCI↓,
Casp↑, chronic and low doses of AEs treatment at sublethal concentrations suppress human breast cancer cell growth via a caspases-independent mechanism.
p16↑, AEs exposure induces a significant increase of senescence-associated β-galactosidase (SA-β-gal) staining and upregulation of tumour suppressor genes, p16INK4a and p21Cip1/Waf1 in MDA-MB231 cells.
P21↑,
TumCCA↑, Cell growth arrest correlates with increased reactive oxygen species (ROS) production in AEs treated breast cancer cells.
ROS↓,
eff↓, Inhibition of ROS generation by N-acetylcysteine (NAC) attenuates the antiproliferative effect.
TumCD↑, After 24 h, 800 μM AEs caused about 50% of death in prostate and melanoma cells; more than 50% of toxicity was detected in breast, ovary, lung, brain, bone, and leukaemia cells.
TumCG↓, After 10-day treatment (from 2.5 μM to 60 μM) direct cell count assay indicated that the cellular growth was significantly inhibited by chronic exposure to low doses of AEs
cellSen↑, AEs Induce Premature Senescence in Breast Cancer Cells
*ROS↓, It is important to highlight that we have previously shown that AEs have a prooxidant activity on breast cancer cells [25] and an antioxidant effect on normal hepatocyte [24].

6896- FIS,    Fisetin is a senotherapeutic that extends health and lifespan
- in-vivo, Nor, NA
*AntiAge↑, Administration of fisetin to wild-type mice late in life restored tissue homeostasis, reduced age-related pathology, and extended median and maximum lifespan.
*cellSen↑, Of the 10 flavonoids tested, fisetin was the most potent senolytic
*selectivity⇅, Fisetin reduced senescence in a subset of cells in murine and human adipose tissue, demonstrating cell-type specificity.

924- RES,    Resveratrol sequentially induces replication and oxidative stresses to drive p53-CXCR2 mediated cellular senescence in cancer cells
- in-vitro, OS, U2OS - in-vitro, Lung, A549
TumCCA↑, S-phase arrest, which is commonly observed in cells treated with RSV
ROS↑,
γH2AX↑, remarkable increase in the amount of γ-H2AX, a marker for DNA double-strand breaks
ATM↑, a master regulator of DNA damage response, was activated by RSV
p‑CHK1↑,
cellSen↑,
CXCR2↑, peaks at day 5 then drops
