Type: Cullin scaffold protein / E3 ubiquitin ligase complex component

Function: Structural scaffold of Cullin-RING ubiquitin ligase 5 (CRL5) complexes that regulate ubiquitination and proteasomal degradation of numerous signaling proteins. CUL5 participates in control of cell proliferation, cytokine and SRC-family signaling, genome stability, cell-cycle regulation, and protein homeostasis.

Cancer: ↕ Context-dependent, but frequently tumor-suppressive. Reduced or lost CUL5 can promote oncogenic signaling, genomic instability, proliferation, and tumor development in several cancers. CUL5 restoration can suppress tumor growth through degradation of oncogenic signaling proteins. However, CUL5 can have pro-tumorigenic functions in some cancer contexts.
