JAG1 (Jagged canonical Notch ligand 1; Jagged1) is a membrane-bound ligand that activates NOTCH-family receptors on adjacent cells. JAG1–Notch signalling regulates cell fate, differentiation, stem-cell maintenance, angiogenesis and communication between tumour cells and the tumour microenvironment. JAG1 is frequently overexpressed in established cancers and can promote cancer-stem-cell properties, proliferation, epithelial–mesenchymal transition, migration, invasion, angiogenesis, metastasis and resistance to anticancer treatment. JAG1 inhibition or downregulation has reduced tumour-cell growth and invasiveness in several preclinical models. The typical cancer-associated direction is up, while the desired anticancer modulation is generally down or blockade of JAG1–Notch signalling.
