ADCY4 (adenylate cyclase 4; adenylyl cyclase 4; AC4) is a membrane-associated enzyme that converts ATP into cyclic AMP (cAMP) in response to G-protein-coupled-receptor signalling. Through cAMP, ADCY4 can influence PKA, CREB, EPAC, calcium regulation, transcription, metabolism, proliferation and differentiation. Altered ADCY4 expression or DNA methylation has been identified in some cancers, including breast cancer, suggesting potential use as a biomarker and signalling regulator. However, current evidence does not establish ADCY4 as a broadly oncogenic or tumour-suppressive target across cancers.
